Abstract

Dori vositalari chaqiradigan gepatotoksiklik (Drug-Induced Liver Injury, DILI) zamonaviy farmakoterapiyaning dolzarb muammolaridan biri hisoblanadi. Jigar organizmning asosiy metabolik va detoksikatsion a’zosi bo‘lgani sababli ko‘plab dori vositalari aynan ushbu organda biotransformatsiyaga uchraydi. Ayrim preparatlar yoki ularning metabolitlari gepatotsitlarga bevosita toksik ta’sir ko‘rsatishi, oksidlovchi stressni kuchaytirishi, mitoxondriya faoliyatini buzishi hamda immun javobni faollashtirishi natijasida jigar shikastlanishi yuzaga keladi. Ushbu maqolada dori vositalari chaqiradigan gepatotoksiklikning asosiy molekulyar mexanizmlari, xavf omillari, klinik ahamiyati hamda profilaktika choralari zamonaviy ilmiy adabiyotlar asosida tahlil qilingan. Shuningdek, farmatsevtlar va tibbiyot xodimlarining gepatotoksik reaksiyalarni erta aniqlash hamda oldini olishdagi o‘rni yoritilgan.
